My nightstand rig is a G23 fitted with a TLR-2 with 1 mag in the well and 1 on the side. As a nightstand rig, I do not keep a round in the chamber.
Personally, when awakened from a dead sleep, I want to have a second to assess the situation prior to racking the slide and going hot. The alarm should afford a decent buffer of warning. If the BG is standing over me, it really will not matter if I have a round in the chamber.
