I just typed in "bulletproof vest" into the search funtion, and came up with 9 pages already answered on this fourm in the past, in reference to your question. Why don't you try that, and then if you have any follow-up questions, we can answer those.

I have been wondering what happens when someone is hit with say a .45 and is wearing a vest? It's gotta hurt! Will it knock you down?

It won't knock you down (it would require a LOT of force to do that). However the impact may make you stagger by surprise and you may lose your balance. If the vest you're wearing can stop the round from penetrating, you'll probably just suffer a bruise from the impact, you might take a broken rib depending upon the force of the impact.

When people get hit and "fall" back or stumble back, it's a bodily reaction to the surprise of being shot. Newton's third laws says, "Whenever a particle A exerts a force on another particle B, B simultaneously exerts a force on A with the same magnitude in the opposite direction." In order for ANY bullet from ANY gun to knock the person being shot down, the gun would have to exert enough force for the shooter to also fall down.

Search youtube for bulletproof there are plenty of videos of people shooting themselves.

Broken ribs and a large bruise that will continue to get bigger.
