Other than the obvious Caliber, what is the difference in a 4506 and 1006, would they fit in same hostlers etc?

A 1006 is the same frame size as the 4506-1 frame.

Second owner. I bought it off a family friend. He only put one magazine through the gun and I myself put less than 100 rounds through the gun.
