As to the bag itself, I recently got one of these--

It can hold a lot of stuff if need be, but is not so large as to be come a beast. I like the way that the two dividers in the main compartment can be configured however you want. One minor caveat is that the shoulder strap is not detachable. In actual use, I find that this is really not an issue.

Being a cheap bastid, for targets I use white paper plates with Post-it notes glue sticked in the center. I staple them square for iron sights, and on diamond for scope cross hair reticles.

OP, Check out some of the gear and set-ups other shooters are using and take note. Some guys bring so much stuff, they spend more time fooling around with crap instead of shooting. After a couple of trips to the range, you'll likely have it figured out. Just some basic things.

Here's my list. My range box is a wheeled pelican case. Documentation refers to stuff like registrations and, now, the statement regarding the injunction against the standard mag ban. Of course, I've got a bunch of other stuff in my truck -- which goes with me always.

Firearms (safe and legal for travel)

                Cooler with iced drinks

                Range mat

                Ammunition

                Range Box

                - Eye protection
                - Ear protection
                - Bench rest
                - Spotting scope
                - Tripod
                - Staple gun
                - Tape
                - Targets and dots
                - Magazines
                - Tool box
                - Cleaning kit
                - Bore cleaner
                - Spray oil
                - Documentation

My range bag is simple for me.

                  Mags
                  Ammo
                  Sight pusher(in case)
                  Tools
                  Eyes and ear pro
                  Bag to collect brass
                  Sticky targets
                  Mag loader

Just got this delivered yesterday, time to change my whole setup:

Before this going from my upstairs apartment to the car with all the different hand gun cases, ammo boxes, rifle bags etc was always annoying. I think with this and a single double gun bag I could easily have a nice range day. Looks like it's very high quality and has marked places for the usuals (glasses, ear protection, magazines, ammo, 4 handguns, etc etc)

The most important thing to have inside a gun bag here is a printed out copy of the California gun laws.

Better to argue the merits in the field than at the police station. Arm yourself before you leave the house.
