In short, yes. Obviously you must be referring to a reproduction because no one would do that to an authentic 1800's revolver. To do it correctly a gun smith would need to cut down the barrel, replace the sight and recrown the barrel. Cost-wise you would probably have more into it than the value of the gun.
Functionally, it might not work out too well. There is a reason most BP revolvers were in the 7 - 8" range. True that there were numerous belly guns, but most were long barreled even by modern standards.Comment

Black powder needs a long barrel for complete burning, otherwise you're just blowing unburned powder out the muzzle.
I have an original 1849 Colt pocket model with a 4'' barrel. The loading lever is about as useless as teats on a boar hog.

I'm looking to do the same thing but haven't got to it yet as I'm still working on a 1911 80% project now.
I've seen conversion cylinders for this but recall the gun and conversion cylinder are more $$, but it might be the same cost if you haven't already purchased all your parts already and then pay to have your barrel shortened.
I've heard you want to stay away from brass frames as well if doing a conversion. One that just has brass trigger guard is ok though.
