Do all those little balls rumble down the barrel 'loose'?

You don't need a special barrel, the rifling does tend to make the shot pattern come out weird. The shot column gets jostled and spreads out really rapidly out of firing.

in my experience they go pop but don't cycle the action. Like others have said don't expect much as they are not very humane unless killing worms from a few feet away!

First, Glocks don't have "normal" rifling. Secondly, I've given up with snakeshot in semi autos. I prefer making my own shotshells for revolvers. I take 3-12 rattlers a year with a snubnosed .38 without any problems. If all I had was a 40 cal, I wouldn't think twice about using shotshells in it.

The lead shot is much softer than your steel barrel and will not hurt it or hurt its rifling in any way.
...and yeah, Glocks don't have "normal rifled barrels" because their polygonal rifling is not the norm...
